چکیده

We describe experimental studies of the deformation of giant lipid bilayer vesicles in shear flow. The experiments are carried out with a counterrotating Couette apparatus. The deformation depends on the mechanical properties of the lipid bilayer, the vesicle radius, and the viscosity of the surrounding Newtonian liquid. We show that the relevant mechanical parameter is the bending rigidity. A simple model has been developed that describes the deformation of a vesicle. This model takes thermal undulations of the bilayer into account. We have obtained a value for the bending rigidity of dimyristoyl-phosphatidylcholine bilayers and its value has been compared with literature data and with results from micropipette aspiration experiments. From the measurements we are able to discriminate between unilamellar and multilamellar vesicles. @S1063-651X~97!12212-7#

Membrane viscosity determined from shear-driven flow in giant vesicles.

The viscosity of lipid bilayer membranes plays an important role in determining the diffusion constant of embedded proteins and the dynamics of membrane deformations, yet it has historically proven very difficult to measure. Here we introduce a new method based on quantification of the large-scale circulation patterns induced inside vesicles adhered to a solid surface and subjected to simple sh...

Nonlocal Bending Analysis of Bilayer Annular/Circular Nano Plates Based on First Order Shear Deformation Theory

In this paper, nonlinear bending analysis of bilayer orthotropic annular/circular graphene sheets is studied based on the nonlocal elasticity theory. The equilibrium equations are derived in terms of generalized displacements and rotations considering the first-order Shear deformation theory (FSDT). The nonlinear governing equations are solved using the differential quadrature method (DQM) whic...

Osmotic shrinkage of giant egg-lecithin vesicles.

Osmotic shrinkage of giant egg-lecithin vesicles was observed by phase-contrast microscopy. The vesicles remained or became spherical when shrinking. Small and thick-walled vesicles formed visible fingers attached to the sphere. The water permeability of the single bilayer was found to be 41 micrometers/s. A variety of observations indicate that osmosis induces a parallel lipid flow between the...

Fluid membranes in hydrodynamic flow fields: Formalism and an application to fluctuating quasi-spherical vesicles in shear flow

The dynamics of a single fluid bilayer membrane in an external hydrodynamic flow field is considered. The deterministic equation of motion for the configuration is derived taking into account both viscous dissipation in the surrounding liquid and local incompressibility of the membrane. For quasispherical vesicles in shear flow, thermal fluctuations can be incorporated in a Langevin-type equati...

Phospholipases a2 from Viperidae snakes: Differences in membranotropic activity between enzymatically active toxin and its inactive isoforms.

We describe the interaction of various phospholipases A2 (PLA2) from snake venoms of the family Viperidae (Macrovipera lebetina obtusa, Vipera ursinii renardi, Bothrops asper) with giant unilamellar vesicles (GUVs) composed of natural brain phospholipids mixture, visualized through fluorescence microscopy. The membrane fluorescent probes 8-anilino-1-naphthalenesulfonicacid (ANS), LAUDRAN and PR...
